是一種PI3K樣的絲氨酸/蘇氨酸激酶,包含有PI3K結(jié)構(gòu)域和Rad3結(jié)構(gòu)域,參與了激活細(xì)胞周期關(guān)卡,控制端粒長(zhǎng)度和應(yīng)答DNA損傷等生物學(xué)過程。 DNA雙鏈斷裂會(huì)導(dǎo)致ATM蛋白激酶的激活。可以被ATM磷酸化的下游靶點(diǎn)包括p53和NBS1蛋白以及組蛋白變體H2AX。通過這種方式, ATM可以影響細(xì)胞周期的過程和DSB修復(fù)以及細(xì)胞凋亡。
taxia telangiectasia mutated (ATM), a serine/threonine protein kinase , is a member of PI3K family proteins. ATM is activated by DNA damage. ATM can phosphorylate many kinase such as p53 and CHK2, resulting in apoptosis and cell cycle arrrest. ATM can be retrieved from nuclear extracts by DNA oligonucleotides or by DNA finity chromatography becasue ATM can bind to DNA. The checkpoint clamp proteins may play the role in modulating the signal (via its processing) or the transmission of the signal. The active PIKK then activates and/or recruits the checkpoint kinases of the Chk1/Chk2 families, which act as effector kinases of the DNA damage pathways. DNA damage can be detected (either directly or after processing) by ATM. In vertebrates, different DNA damage signals elicit either a branch composed of the ATM-Chk2 axis.
